Assume the start date and time is in A1 and the end date and time is in B1, put this formula in C1 and format the cell as [hh]:mm

=NETWORKDAYS(A1,B1)-1-MOD(A1,1)+MOD(B1,1)

Using the numbers in your example this formula returns 21:00
