In the attached, the range A1:E30 has been conditionally formatted to turn yellow when a date which is a multiple of 6 days away from the date in row 1 of each column is entered into it.
The conditional format formula is:

=AND(MOD(A1-A$1+1,6)=1,COUNTBLANK(A1)=0)
