Is there a way to change the font size of one line only? For example, if one chapter title is a bit longer and I wish to make the font of that line smaller, can I do so without changing the font size of the entire TOC?

Yes, modify the TOC Style for that particular line to no update automatically. I advise against doing this. I would recommend creating a new style instead and applying that.
